Arrowhead's plozasiran cuts acute pancreatitis event rates 78% versus placebo in pooled trials
ARWR•Arrowhead Pharmaceuticals said pooled SHASTA-3/4 results showed plozasiran reduced acute pancreatitis event rates by 78% versus placebo in severe hypertriglyceridemia. The company also reported three-year triglyceride reductions in a PALISADE extension and early cholesterol and triglyceride reductions with ARO-DIMER-PA.
1. Plozasiran trial results
Arrowhead Pharmaceuticals presented clinical results for plozasiran and ARO-DIMER-PA at the American Heart Association Scientific Sessions 2026. Pooled SHASTA-3/4 results showed plozasiran reduced acute pancreatitis event rates by 78% versus placebo in severe hypertriglyceridemia, particularly among patients with recent pancreatitis.
2. Additional study findings
The PALISADE extension showed sustained triglyceride reductions through three years in familial chylomicronemia syndrome, but had no concurrent placebo control. Early-stage ARO-DIMER-PA results showed simultaneous LDL cholesterol and triglyceride reductions after single doses; those changes were not placebo-adjusted. Injection-site redness was common, and no serious adverse events occurred. Longer-term safety and potential cardiovascular benefits require further study.
