Calidi Demonstrates RedTail-Mediated TROP-2 Engager and IL-15 SA Expression in Tumors
CLDI•Calidi presented preclinical data showing its RedTail virotherapy platform can express high concentrations of a TROP-2-targeting T-cell engager and IL-15 superagonist in the tumor microenvironment, overcoming solid tumor delivery challenges. The company expects to file an IND for its IL-15-engineered lead candidate CLD-401 by year-end 2026.
1. Summit Presentation
At the T-Cell Engager Therapeutic Summit, Calidi showcased data on CLD-501 demonstrating simultaneous in situ expression of a TROP-2-targeting engager and IL-15 superagonist. The results highlight tumor-specific activation of T-cells and remodeling of the tumor microenvironment to enable solid tumor targeting.
2. RedTail Platform Technology
The engineered oncolytic virus selectively homes to tumors after systemic delivery, limiting peripheral exposure while driving high-level genetic payload expression in situ. Its dual expression capability addresses historical barriers to T-cell engager efficacy in solid tumors by combining immune activation with localized engager production.
3. Clinical Pipeline and Timeline
Calidi plans to submit an IND application for CLD-401, its IL-15 superagonist candidate, by the end of 2026. The company is also evaluating additional T-cell engager targets including EGFR, EpCAM and Nectin-4 to expand its solid tumor pipeline.
