Celldex discontinued its Phase 2 prurigo nodularis trial of barzolvolimab after missing the primary endpoint at Week 12.
- Key secondary endpoints also missed.
- Results did not differentiate from placebo.
- No improvement was seen through Week 24.
- The safety profile remained favorable.
- The new 450 mg loading dose followed by 150 mg or 300 mg every four weeks was well tolerated.
- Serum tryptase dropped sharply, signaling systemic mast cell depletion. The biomarker effect did not translate into symptom improvement in prurigo nodularis.
- Phase 3 chronic spontaneous urticaria topline data are expected in September or October 2026.
