Immuneering Phase 2a Shows 17.3-Month Median Survival with Favorable Tolerability in Pancreatic Cancer
IMRX•Immuneering’s Phase 2a trial of atebimetinib plus modified gemcitabine/nab-paclitaxel in 55 first-line metastatic pancreatic cancer patients showed a 17.3-month median overall survival versus 8.5 months standard of care. Only two grade 3+ treatment-related adverse event categories occurred in ≥10% of patients, and 84% maintained or gained weight at three months.
1. Phase 2a Efficacy Results
In the expanded cohort of 55 first-line metastatic pancreatic ductal adenocarcinoma patients, the combination of atebimetinib with modified gemcitabine/nab-paclitaxel achieved a median overall survival of 17.3 months (95% CI: 11.2, not reached) versus 8.5 months median OS in the historical MPACT study. Median progression-free survival was 8.3 months (95% CI: 5.9, 9.6), with a disease control rate of 82% and a confirmed overall response rate of 36%.
2. Safety and Tolerability Findings
Only two categories of Grade 3 or higher treatment-related adverse events occurred in at least 10% of patients, both attributed to chemotherapy, with no Grade 4 or 5 events related to atebimetinib. Additionally, 84% of participants maintained or gained weight at three months, suggesting preserved performance status and potential mitigation of cachexia.
3. Phase 3 MAPKeeper 301 and Next Milestones
Immuneering is actively recruiting for the global randomized Phase 3 MAPKeeper 301 trial comparing atebimetinib plus mGnP versus standard gemcitabine/nab-paclitaxel, with first patient dosing expected in mid-2026. The company will host an investor conference call on June 1, 2026, at 8:00 a.m. EDT to discuss these clinical updates.
