Sagimet’s Denifanstat/Resmetirom Combo Cuts LDL by Up to 17mg/dl
SGMT•Sagimet Biosciences' Phase 1 trial of denifanstat and resmetirom in 40 healthy adults demonstrated rapid total cholesterol reductions of 23mg/dl and LDL decreases of 17mg/dl in one cohort and 16mg/dl and 14mg/dl respectively in another. The combo was selected as a Best of EASL 2026 presentation.

Sagimet Biosciences presented two posters at the European Association for the Study of the Liver Annual Congress 2026 in Barcelona, including its denifanstat and resmetirom combination therapy chosen as a Best of EASL presentation for rapid lipid reductions in healthy adults.
2. Phase 1 Clinical Data
An open-label trial in 40 healthy adults (mean BMI 30–32 kg/m2) evaluated denifanstat 50mg and weight-based resmetirom (80/100mg) over two seven-day monotherapy followed by seven-day combination periods, reporting total cholesterol drops of 23mg/dl (cohort 1) and 16mg/dl (cohort 2), LDL decreases of 17mg/dl and 14mg/dl, and no safety signals.
3. Preclinical Mechanistic Insights
In an LDL receptor knockout mouse model of MASH, the denifanstat/resmetirom combo outperformed monotherapy in liver histology and lipid lowering, with transcriptomic analysis revealing enhanced anti-inflammatory, anti-fibrotic activity and improved regulation of lipid and cholesterol synthesis pathways.
