Elraglusib Plus Chemo Extends mOS to 16.9 Months in KRAS WT Pancreatic Cancer
ACTU•Actuate Therapeutics posted Phase 2 data showing elraglusib plus gemcitabine/nab-paclitaxel extended median overall survival to 16.9 months in KRAS wild-type mPDAC patients versus 10.1 months with chemotherapy alone, and TP53 wild-type patients saw 13.4 vs 7.6 months. Elraglusib combinations with FOLFIRINOX achieved 9.8 months mOS and 6.5 months mPFS versus 7.7 and 5.1 months, respectively, with standalone FOLFIRINOX and were well tolerated with 34.7% grade 3+ adverse events.
1. ASCO Poster Presents Phase 2 GnP Data
Actuate Therapeutics presented post-hoc efficacy and biomarker results from its randomized Phase 2 study in previously untreated metastatic pancreatic ductal adenocarcinoma patients, showing elraglusib combined with gemcitabine/nab-paclitaxel extended median overall survival compared with chemotherapy alone.
2. Biomarker and Subgroup Analyses
KRAS wild-type patients achieved median overall survival of 16.9 versus 10.1 months and TP53 wild-type patients 13.4 versus 7.6 months; exploratory analyses also linked ECOG performance status, albumin levels, CA19-9, tumor grade and ctDNA fraction to survival benefits only in the elraglusib arm.
3. Phase 2 FOLFIRINOX Combination Results
In a separate Phase 2 study, elraglusib combined with FOLFIRINOX, with or without losartan, delivered median overall survival of 9.8 months and median progression-free survival of up to 6.5 months versus 7.7 and 5.1 months with FOLFIRINOX alone, with 34.7% of patients experiencing grade 3 or higher adverse events.
