The U.S. Patent and Trademark Office issued a Notice of Allowance for Hoth’s HT-KIT platform, granting composition-of-matter and method claims for antisense oligomers of 25–50 nucleosides directed at MS4A6A pre-mRNA splicing. These claims include pharmaceutical compositions and methods to reduce FcεRI surface expression by exon-skipping in mast cells.
Allowed method claims span eight mast cell–driven indications: asthma, atopic dermatitis, chronic rhinitis, allergic conjunctivitis, chronic sinusitis, anaphylaxis prevention, mastocytosis and mast cell tumors. This broad coverage enhances potential market reach and competitive positioning for HT-KIT.
The allowance establishes a defensible IP foundation for the lead antisense oligomer (SEQ ID NO: 22) and its modified embodiments, reinforcing HT-KIT’s orphan drug status. Hoth plans to finalize its IND in 2026, leveraging this strengthened IP to advance first-in-human studies and explore dual-target antisense combinations.
