Altimmune’s Pemvidutide Cuts Triglycerides 23.7%, Yields 7.5% Weight Loss at 48 Weeks
ALT•Altimmune’s 48-week IMPACT Phase 2b data show pemvidutide 1.8 mg reduced triglycerides by 23.7% and total cholesterol by 15.4%, while producing 7.5% weight loss and lowering systolic blood pressure by 4.0 mmHg in MASH patients. Trial reported only 1% discontinuations due to adverse events, supporting a favorable safety profile.
1. Study Design and Patient Population
The randomized, double-blind IMPACT Phase 2b trial enrolled 212 biopsy-confirmed MASH patients with fibrosis stages F2 or F3. Participants were assigned 1:2:2 to weekly subcutaneous pemvidutide at 1.2 mg, 1.8 mg or placebo for 48 weeks to evaluate liver fat, fibrosis biomarkers and metabolic endpoints.
2. Metabolic Improvements
Pemvidutide 1.8 mg achieved a 23.7% reduction in triglycerides and 15.4% decrease in total cholesterol versus placebo. Additional benefits included 7.5% weight loss, a 3.0 kg/m2 BMI reduction, 5.3 cm smaller waist circumference and 4.0 mmHg lower systolic blood pressure.
3. Safety Profile
Treatment discontinuations due to adverse events were limited to 1% of pemvidutide patients. Gastrointestinal events were mostly mild to moderate and occurred primarily within the first 8 weeks, with no increase in cardiac adverse events compared to placebo.
4. Phase 3 PERFORMA Trial Plans
Altimmune plans to initiate the multinational, randomized PERFORMA Phase 3 trial in MASH patients in the second half of 2026 to confirm the efficacy and safety of pemvidutide across liver and metabolic endpoints.
